WHAT IS BIOTICAL CRYPTO+GIARDIA CARD?biotical Crypto+Giardia card is a colored chromatographic immunoassay test for the simultaneous qua-litative detection of Cryptosporidium and Giardia antigens in stool samples. Thanks to its easy handling and high sensitivity, the healthcare professional will be able to make a presumptive diagnosis of criptos-porosis and/or Giardiasis.

CRYPTOSPOROSIS AND GIARDIASISGiardia and Cryptosporidium parvum are the most frequent parasites that cause water-borne diseases in humans: cryptosporidiosis and Giardiasis.

Giardiasis is an intestinal disease caused by Giardia intestinalis (also called Giardia lamblia and Giardia duodenalis), a single-celled microscopic parasite that is found in the intestine of people and animals and is transmitted through the feces of an infected person or animal.

This parasite is protected by an outer covering (cyst) that allows them to survive outside the body and in the environment for long periods of time. Its infective form is usually by fecal-oral transmission. Among the most common symptoms of both diseases are: watery diarrhea, cramps and stomach disorders. In immunocompromised people or in elderly people, complications can appear that lead to serious illness and even death.

Cryptosporidium parvum is the most frequent cause of diarrheal processes in developing countries. This parasite is in the majority of enteric pathogenic infections, being transmitted feco-orally. Some of the most common symptoms of cryptosporidiosis are watery diarrhea, severe stomach pains, weight loss, nausea, and sometimes fever.

TEST LINES (T1 / T2):If the sample is positive:

The antigens of the patient’s stool sample will react with one (or both) of the red colored conjugate complexes (anti- Cryptosporidium monoclonal antibodies - latex red microspheres for the line T1 and anti- Giardia monoclonal antibodies - red latex microspheres for the line T2) and it will advance by ca-pillary through the membrane. As the sample migrates, so do the conjugate complexes. The antibodies anti- Cryptosporidium present in the T1 line and / or the antibodies anti- Giardia present in the T2 line will capture the complex giving rise in a red line.

If the sample is negative:If the sample is negative and therefore does not contain antigens (or the antigen is present in a concen-tration below the detection limit), there is no reaction with the red colored complexes. The anti- Cryp-tosporidium and anti- Giardia antibodies present in the T1 and T2 line will not capture the red colored antigen-complex (not formed) and the red lines will not appear.

CONTROL LINE (C):Regardless of whether the sample is positive or not, to verify that the test has been performed correctly

STORAGE AND STABILITY Store as packaged in the sealed pouch at 2-30 ºC. The test is stable until the expiration date marked on its sealed pouch. The test must remain in the sealed pouch until use. Do not freeze.

PRECAUTIONSFor professional in vitro diagnostic use.Do not use after expiration date.All the specimens should be considered potentially hazardous and handled in the same manner as an infectious agent. A new test must be used for each sample to avoid contamination errors.The test should be discarded in a proper biohazard container after testing.Reagents contain preservatives. SPECIMEN COLLECTION AND PREPARATIONStool samples should be collected in clean containers. The samples can be stored in the refrigerator (2-8 ºC) for 1-2 days prior to testing. For longer storage, maximum 1 year, the specimen must be kept frozen at -20 ºC. In this case, the sample will be totally thawed and brought to room temperature before testing. Homogenise stool sample as thoroughly as possible prior to preparation.

Specimen preparation (see illustration):

Then, introduce the stick once into 4 different parts of the stool sample (2), to collect faecal sample (approx. 125mg) and add it to the stool collection tube. For liquid samples, add approx. 125µL in the stool collection tube using a micropipette.Close the tube with the diluent and stool sample. Shake the tube in order to assure good sample dispersion (3).

VERY IMPORTANT:

TEST PROCEDURE

Proceed to shake the stool collection tube in order to assure good sample dispersion. Remove biotical Crypto+Giardia card test from its sealed bag just before using it. Take the stool collection tube, cut the end of the cap (4) and dispense 4 drops in the circular window marked with the letter A (5) and 4 drops, using the same tube, in the circular window marked with the letter B (6). Avoid adding solid particles with the liquid.

If the test does not run due to solid particles, stir the sample added in the sample window (S) with the stick. If it doesn´t work, dispense a drop of diluent until seeing the liquid running through the reaction zone.

NOTES ON THE INTERPRETATION OF RESULTSThe intensity of the red coloured bands in the test lines (T) in the results windows will vary depending on the concentration of antigens present in the specimen. However, neither the quantitative value nor the rate of increase in antigens can be determined by this qualitative test.

LIMITATIONS The test must be performed within 2 hours after opening the sealed pouch.

Neither the quantitative value nor the rate of increase in Cryptosporidium and/or Giardia antigens concentration can be determined by this test.An excess of sample could cause wrong results (brown bands appear). Dilute the sample with the buffer and repeat the test.Samples should been collected as soon as possible once the symptoms appear. After one week of infection the number of parasites decrease, making the sample less reactive.Some stool samples can decrease the intensity of the control line.If the test result is negative and clinical symptoms persist, additional testing using other clinical methods are recommended. CRIPTOSPOROSIS Y GIARDIASISGiardia y Cryptosporidium son los parásitos más frecuentes causantes de enfermedades transmitidas a través del agua en humanos: la criptosporidiosis y la Giardiasis.

La Giardiasis es una enfermedad intestinal ocasionada por Giardia intestinalis (llamado también Giardia lamblia y Giardia duodenalis), un parásito microscópico unicelular que se encuentra en el intestino de las personas y los animales y se transmite a través de las heces de una persona o animal infectado.

LÍNEAS DE TEST (T1 / T2):Si la muestra es positiva: Los antígenos de la muestra de heces del paciente reaccionarán con alguno (o con ambos) de los complejos conjugados coloreados rojos (anticuerpos monoclonales anti- Cryptosporidium - microesferas rojas de látex para la línea T1 y anticuerpos monoclonales anti- Giardia -microesferas rojas de látex para la línea T2) y avanzarán por capilaridad a través de la membrana. Conforme la muestra va migrando también lo hacen los complejos conjugados. Los anticuerpos anti- Cryptosporidium presentes en la línea de T1 y/o los anticuerpos anti- Giardia presentes en la línea de T2 capturarán el complejo dando lugar a una línea roja.Si la muestra es negativa: Si la muestra es negativa y por lo tanto no contiene antígenos (o estos está presentes en una concentración inferior al límite de detección), no se producirá reacción con los complejos coloreados rojos. Los anticuerpos anti-Giardia y anti-Crypto presentes en las líneas T1 y T2 no capturarán los antígenos-complejos coloreados y no aparecerán las líneas rojas.

No se deben usar con componentes de otros kits comercializados.Seguir las Buenas Prácticas de Laboratorio, llevar ropa de protección adecuada, usar guantes desechables, gafas de protección y mascarilla. No comer, ni beber o fumar en la zona de trabajo.Es posible que aparezcan bandas color amarillo o naranja pálido en la ventana de resultados de las tiras de test, en las posiciones donde deberán aparecer las bandas coloreadas de test y control, antes de que se realice la prueba. Estas bandas son completamente normales y corresponde a los anticuerpos de captura de la prueba. La existencia de estas bandas no alteran el estado de conservación y funcionalidad del kit.

RECOGIDA DE MUESTRAS Y PREPARACIÓNLas muestras deben ser recogidas en un recipiente limpio. Las muestras se deben conservar en frío (2-8 ºC), durante 1-2 días, hasta el momento de utilizarlas. Para conservar las muestras durante un tiempo prolongado, como máximo 1 año, deben mantenerse congeladas a -20 ºC. En este caso, la muestra debe descongelarse totalmente y alcanzar la temperatura ambiente para poder utilizarla en la prueba. Homogenizar la muestra vigorosamente antes de su preparación.

Preparación de la muestra (ver dibujo):

PROCEDIMIENTOPreviamente los test, las muestras y los controles se deben acondicionar a la temperatura

Agitar el tubo para dilución de muestra para asegurar una buena dispersión. Sacar el test CerTest Crypto+Giardia de su envase antes de utilizarlo.Tomar el tubo para dilución de muestra, cortar la punta del tapón (4) y añadir 4 gotas del líquido en la ventana circular marcada con la letra S (5) evitando añadir partículas sólidas con el líquido.

Si se da el caso de que el test no funciona debido a la presencia de partículas sólidas, agitar con el palito la muestra en la ventana (S). Si no funciona, añadir una gota de diluyente hasta que se vea avanzar el líquido por la zona de resultados.

LIMITACIONES El test debe ser realizado como máximo hasta 2 horas después de haber abierto la bolsa que lo contiene.Solo se pueden analizar muestras de heces frescas o congeladas sin conservar.biotical Crypto+Giardia card solo indicará la presencia de Cryptosporidium y/o Giardia en la muestra (detección cualitativa) y debe usarse solamente para la detección de antígenos de Cryptosporidium y/o Giardia en muestras de heces humanas. Esta prueba no puede determinar ni el valor cuantitativo ni la tasa de aumento en la concentración de antígenos de Cryptosporidium y/o Giardia.Un exceso de muestra puede causar resultados incorrectos (aparecen bandas marrones). En este caso diluya la muestra con el tampón y repita la prueba.Las muestras deben recogerse lo antes posible una vez que aparezcan los síntomas. Después de una semana de infección, la cantidad de parásitos disminuye, lo que hace que la muestra sea menos reactiva.Algunas muestras de heces pueden disminuir la intensidad de la línea de control.Si el resultado de la prueba es negativo y los síntomas clínicos persisten, se recomiendan pruebas
